Unlocking the Trial of the Sekhemas:
To begin, defeat Balbala the Traitor, located in Traitor's Passage within Act 2. Balbala presents a formidable challenge early in the game due to her swift and powerful attacks. Upon defeating her, she drops Balbala's Barya, the key item needed to access the Trial.
After this victory, travel to the Trial of the Sekhemas location using the Ardura's travel map or waypoint. You'll find a ruined temple where Balbala, now acting as a guide, awaits. Place Balbala's Barya on the Relic Altar to initiate your Trial run.
